Producció i estudi funcional de la proteïna recombinant DFV-D1

Abstract

Aquest treball presenta els resultats d’un projecte de desenvolupament de dos compostos amb interès comercial i que s’emmarca dintre del conveni de transferència de coneixement establert entre la UVic-UCC i Divasa-FarmaVic (DFV). Concretament es mostren els resultats de l’estudi de diferents propietats funcionals que presenta la proteïna recombinant DFV-D1, propietat de DFV, així com la manera d’obtenir-la a escala de laboratori i amb la puresa suficient com per dur a terme assajos posteriors. Paral•lelament també s’ha avaluat el compost DFV-M1 per a unes propietats funcionals d’interès i la manca d’altres propietats indesitjades. L’objectiu principal d’aquest treball és avaluar experimentalment i mitjançant el mètode científic, dues activitats diferents que presenta DFV-D1 i el mateix per una activitat de DFV-M1 per les quals aquests compostos es presenten interessants per a incloure’ls com a ingredients en determinats productes fitosanitaris que comercialitza DFV. Per dur a terme aquests objectius, primer de tot s’ha hagut de produir i purificar DFV-D1, si bé és cert que també s’ha intentat purificar a partir de material encara no processat que ha estat proporcionat per la mateixa empresa. Un cop DFV-D1 ha estat purificada s’han emprat tècniques de laboratori i kits comercials per tal d’avaluar-ne les activitats funcionals. En el cas de DFV-M1 no ha estat necessari de produir-la ja que també ha estat proporcionada per DFV. Durant el projecte s’han desenvolupat satisfactòriament dues noves tècniques reproduïbles que permeten avaluar les propietats funcionals dels mencionats compostos. A més s’han pogut realitzar altres assajos amb tècniques prèviament desenvolupades i que són propietat de DFV per avaluar-ne altres característiques d’interès. Tot i que el volum de dades obtingut no ha estat suficient per dur a terme un tractament estadístic d’aquest els resultats han estat bons i són prometedors de cara a una continuïtat del projecte.

This assay shows the results from a development project of two compounds with commercial interest within the agreement of knowledge transfer between UVic-UCC and Divasa-FarmaVic (DFV). Specifically, here it is shown the results of the study of different functional properties that features the recombinant protein DFV-D1, which is property of DFV, and the method to obtain it at a laboratory scale with a proper purity for the correct perform of further experiments. At the same time, the other compound DFV-M1 has been evaluated for other functional properties of interest as well as for the lack of other undesired properties. The main objective of this work is to evaluate experimentally and through the scientific method, two different activities that performs DFV-D1, as well as another activity that posses DFV-M1, for which these compounds are of great interest and could be included as ingredients for different phytosanitary products that DFV commercialize. To achieve this goal, first of all it was needed to produce and purify DFV-D1, although it was also intended to purify it from unprocessed material subministrated by the company. As soon as DFV-D1 has been purified, different laboratory techniques and commercials kits have been used to evaluate the different functional activities. In the DFV-M1 case, it has not been necessary to produce and purify it before the activity assays because it has been provided by DFV and already purified. During this project, two new reproducible techniques that allow to evaluate the functional properties of the mentioned compounds have been developed. Furthermore, other assays have been conducted with previosly developed techniques which are property of DFV to evaluate other characteristics of interest. Although the volum of data obtained has not been suficient to perform a statistical analisys the results have been good and promising in a way that the project could continue in a near future.
