Caracterització proteòmica dels subtipus de carcinoma colorectal en humà en model PDX

Abstract

En aquest Treball de Fi de Grau s’exposen els resultats de la caracterització proteòmica dels diferents subtipus de càncer colorectal (MSS i MSI) a partir de mostres PDX obtingudes del laboratori del Dr. H. G. Palmer (Puig I. et al., 2013). El propòsit de l'estudi ha sigut la posta a punt de la metodologia de l’anàlisi proteòmica, dirigida a contrastar la classificació de CRCs proposta en la bibliografia, i en particular la recerca de marcadors per a cada subtipus de CRC. Una de les principals avantatges de l'estudi plantejat, deriva del fet que el disposar dels models PDX, permetrà contrastar de forma eficient en assajos pre-clínics les hipòtesis derivades de l'anàlisi proteòmica, ja sigui pel que fa a possibles marcadors de resposta, resistència, o dianes terapèutiques específiques dels diferents subtipus de tumor de còlon identificats. Els resultats del projecte han permès establir un workflow de treball òptim per a l’obtenció d’extractes proteics i digerits tríptics en mostres PDX i la seva anàlisi proteòmica. Els resultats d’aquesta anàlisi han permès la correcta classificació de les mostres estudiades en els corresponents subtipus. Entre les proteïnes diferencials estudiades, s’ha identificat TCF7L1, TCF7L2, LEF1, TCF7, CTNNBIP1, MAPK1 i PEA15, que activen vies de senyalització específiques pel subtipus MSS i MPI, GMMPA i GMPPB, específiques pel subtipus MSI. Per l’activitat de MAPK1 s’han realitzat experiments de validació per Western Blot i immunohistoquímica. Finalment s’han comparat els subtipus de CRC MSS i MSI contra una metàstasi de càncer corioembrionàri (PD) a colon i s’han pogut observar les diferencies dels proteomes. Aquest ha sigut un bon punt de partida per a la continuïtat del projecte amb un nombre de mostres molt més elevat que permetrà obtenir un gran poder estadístic per a la recerca de marcadors terapèutics tant per MSS com per MSI.

This dissertation exposes the results of proteomic characterization of the different subtypes of colorectal cancer (MSS and MSI) from PDX samples obtained from the laboratory of Dr. H. G. Palmer (Puig I. et al., 2013). The purpose of the study was to establish a proteomic analysis methodology, aimed at comparing the CRC proposed classification in the literature, and to find molecular markers for each CRC subtype. One of the main advantages of the study is the fact to work with PDX models, that can allow testing of pre-clinical hypotheses derived from the proteomic analysis. Project results have enabled to establish an optimal workflow for preparation and proteomic analysis of PDX samples. The results of this analysis have allowed us to classify our samples in the relevant subtypes. Among the differential proteins found in the study were TCF7L1, TCF7L2, LEF1, TCF7, CTNNBIP1, MAPK1 and PEA15, identified as activators of specific signaling pathways. MPI, GMPPB and GMMPA have been found to be increased for MSI subtype. The observed MAPK1 activation found in MSS subtype has been validated through Western Blot and immunohistochemistry experiments. Finally, we have comparison of MSS and MSI CRC subtypes against a cancer metastasis sample of coriembrionary origin (PD), has demonstrated large differences at proteomic level. This work constitutes a good starting point for the continuation of the project with a larger number of samples which will provide higher statistical power to search therapeutic research markers of the different CRC subtypes.
