Al llarg de la vida, estem exposats a diversos compostos químics que estan presents en l’aire,
aigua, o en els aliments. Aquests compostos poden interactuar amb la cèl·lula mitjançant les
proteïnes de membrana i causar desregulacions en la funció de les cèl·lules.
En els últims anys el mètode utilitzat per poder identificar aquests proteïnes diana ha estat el mètode PISA que es basa en el tractament tèrmic per induir la precipitació de les proteïnes.
En aquest projecte, el proteoma de membrana extret de teixit de fetge has estat solubilitzat utilitzant un detergent. A més a més, s’ha validat un nou mètode basat en el mètode PISA, que ha estat modificat per tal d’ evitar un mètode basat en el tractament tèrmic. Específicament,
s’utilitzen ions de Zn2+ per d’induir la precipitació de proteïnes. Al llarg de la validació varis
factors han estat avaluats. En concret quina és la correcte centrifugació, la concentració de Zn2+
necessària o el temps que es necessita per incubar les mostres.
S’ha pogut demostrar que les proteïnes de membrana es poden solubilitzar amb Igepal CA-630.
També, a mesura que es validava el nou mètode s’ha observat que per obtenir les proteïnes precipitades, aquetes han d’estar incubades amb ions de Zn2+ a una concentració entre 750 i 1000 μM i incubades per 30 minuts. Un cop incubades han de ser centrifugades a 17 000 g per 10 minuts.
Aquest projecte demostra que el nou mètode basant en l’ús de ions de Zn2+ a l’hora d’induir
precipitació és efectiu quan es treballa amb proteïnes solubilitzades.^^^^

Throughout life we are exposed to several chemical compounds present in the air, water, or
food. By being exposed to those, some proteins can be targeted by them, leading cell functional
dysregulations. In the recent years PISA method, which is based on the use of thermal approach
to induce proteins precipitation. In this project membrane proteome extracted from liver tissue, was solubilized with the use of detergents. Moreover, a new method based on PISA method but modified to use a non-isothermal approach was validated. Specifically, we used Zn2+ to induce precipitation. During the validation of this new method, some factors as which centrifugation
should be used, how much time samples should be incubated with Zn2+ ions and which is the
concentration necessary for Zn2+ were evaluated.
It was demonstrated that membrane proteome could be successfully solubilized by using detergent Igepal CA-630. Also, while validating the new method it was shown that proteins
needed to be incubated for 30 minutes at a Zn2+ concentration between 750 and 1000 μM. Once incubated they should be centrifuge at 17 000 g for 10 minutes.
In conclusion, it has been demonstrated that using Zn2+ ions in order to induce precipitation is effective when working with solubilized proteome.^^^^
